Dietary fatty acids intake affects the composition of erythrocyte fatty acids, which is strongly correlated with glycolipid metabolism disorders. This study aimed at investigating the different effects of marine-derived and plant-derived omega-3 polyunsaturated fatty acid (n-3 PUFA) on the fatty acids of erythrocytes and glycolipid metabolism in patients with type 2 diabetes mellitus (T2DM). The randomized double-blinded trial that was performed on 180 T2DM patients. The participants were randomly assigned to three groups for the six-month intervention. The fish oil (FO) group was administered with FO at a dose of 3 g/day containing eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), the perilla oil (PO) group was administered with PO at a dose of 3 g/day containing ¦Á-linolenic (ALA), the linseed and fish oil (LFO) group was administered with mixed linseed and fish oil at a dose of 3 g/day containing EPA, DHA and ALA. Demog. information were collected and anthropometric indexes, glucose and lipid metabolism indexes, erythrocyte fatty acid composition were measured. Statistical analyses were performed using two-way ANOVA. A total of 150 patients finished the trial, with 52 of them in the FO group, 50 in the PO group and 48 in the LFO group. There were significant effects of time x treatment interaction on fast blood glucose (FBG), insulin, HOMA-IR and C-peptide, TC and triglyceride (TG) levels (P < 0.001). Glucose and C-peptide in PO and LFO groups decreased significantly and serum TG in FO group significantly decreased (P < 0.001) after the intervention. Erythrocyte C22: 5 n-6, ALA, DPA, n-6/n-3 PUFA, AA/EPA levels in the PO group were significantly higher than FO and LFO groups, while EPA, total n-3 PUFA and Omega-3 index were significantly higher in the FO and LFO groups compared to PO group. Supplementation with perilla oil decreased FBG while fish oil supplementation decreased the TG level. Marine-based and plant-based n-3 PUFAs exhibit different effects on fatty acid compositions of erythrocytes and regulated glycolipid metabolism This trial was recorded under Chinese Clin. In the breeding process of crispy grass carp, the addition of FB is very high, which has adverse effects on the physiol. function of grass carp. In order to determine the optimum addition amount of FB in crispy grass carp culture, five isonitrogenous (approx. 33.79% crude protein, CP) and isolipidic (approx. 3.43% crude lipid) diets containing different levels of FBM replacing protein materials 0% (CON), 70% (FBM70), 80% (FBM80), 90% (FBM90) and 100% (FBM100) were formulated and were randomly assigned to triplicate groups of fish for 100 days. The results showed that FBM supplement significantly enhanced the myofiber area and radius, content of collagen and CP in muscle. Moreover, compared with the CON group, the content of inosine 5¡ä-monphosphate (IMP), total amino acid (TAA), total non-essential amino acid (TNAA) and the expression of collagen-related (collagen type 1 alpha 1 chain [col1a1] and collagen type 1 alpha 2 chain [col1a2]) and muscle quality-related genes (fibroblast growth factor 6a [fgf6aI] and fibroblast growth factor 6b [fgf6b]) was the highest in the FBM70 group and the lowest in the FBM100 group. In conclusion, the maximal proportion of dietary protein replacement with FBM was 70% based on the flesh quality of grass carp, which provided guidance for the cultivation of crisp grass carp.

Synovial fluid (SF) is commonly used for diagnostic and research purposes, as it is believed to reflect the local inflammatory environment. Owing to its complex composition and especially the presence of hyaluronic acid, SF is usually viscous and non-homogeneous. In this study, we investigated the importance of homogenization of the total SF sample before subsequent anal. SF was obtained from the knee of 29 arthritis patients (26 rheumatoid arthritis, 2 osteoarthritis, and 1 juvenile idiopathic arthritis patient) as part of standard clin. care. Synovial fluid was either treated with hyaluronidase as a whole or after aliquoting to determine whether the concentration of soluble mediators is evenly distributed in the viscous synovial fluid. Cytokine and IgG levels were measured by ELISA or Luminex and a total of seven fatty acid and oxylipin levels were determined using LC-MS/MS in all aliquots. For cell anal., synovial fluid was first centrifuged and the pellet was separated from the fluid. The fluid was subsequently treated with hyaluronidase and centrifuged to isolate remaining cells. Cell numbers and phenotype were determined using flow cytometry. In all patients, there was less variation in IgG, 17-HDHA, leukotriene B4 (LTB4), and prostaglandin E2 (PGE2) levels when homogenization was performed before aliquoting the SF sample. There was no difference in variation for cytokines, 15-HETE, and fatty acids arachidonic acid (AA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA). Between 0.8 and 70% of immune cells (median 5%) remained in suspension and were missing in subsequent analyses when the cells were isolated from untreated SF. This percentage was higher for T and B cells: 7-85% (median 22%) and 7-88% (median 23%), resp. Homogenization of the entire SF sample leads to less variability in IgG and oxylipin levels and prevents erroneous conclusions based on incomplete isolation of synovial fluid cells.

The COVID-19 pandemic has forced lockdowns and declarations of states of emergency, resulting in marked changes to daily life such as dietary habits in many countries. Though serum omega-3 polyunsaturated fatty acids levels have been shown to be useful markers for recurrent vascular events or worse prognosis in cardiovascular diseases and ischemic stroke, the relationship between serum omega-3 PUFA levels and the occurrence of intracerebral hemorrhage has essentially been unknown. We explored the association of serum omega-3 polyunsaturated fatty acids with intracerebral hemorrhage during the COVID-19 pandemic. Participants comprised patients admitted to Juntendo University Hospital (Tokyo, Japan) with intracerebral hemorrhage between Jan. 1, 2016 and Apr. 30, 2020. Clin. characteristics including serum omega-3 polyunsaturated fatty acid levels were compared between patients developing intracerebral hemorrhage during the period from Jan. 1, 2016 to Feb. 29, 2020, and the subsequent COVID-19 pandemic period (March 1 to Apr. 30, 2020). Clin. characteristics independently related to intracerebral hemorrhage during the COVID-19 pandemic were analyzed by comparing these two cohorts of intracerebral hemorrhage patients in different periods. A total of 103 patients (age, 67.0 ¡À 13.9 years; 67 males) with intracerebral hemorrhage were enrolled. Intracerebral hemorrhage developed in 91 patients before and 12 patients during the COVID-19 pandemic. Monthly averages of intracerebral hemorrhage patients admitted to our hospital during and before the COVID-19 pandemic were 6 and 1.82, resp. Serum eicosapentaenoic acid levels were significantly lower in intracerebral hemorrhage patients during the COVID-19 pandemic than before (31.87 ¡À 12.93¦Ìg/mL vs. 63.74 ¡À 43.29¦Ìg/mL, p = 0.007). Multiple logistic regression anal. showed that, compared to before the COVID-19 pandemic, dyslipidemia (odds ratio 0.163, 95% confidence interval 0.031-0.852; p = 0.032) and eicosapentaenoic acid levels (odds ratio 0.947, 95% confidence interval 0.901-0.994; p = 0.029) were associated with intracerebral hemorrhage during the COVID-19 pandemic. From our preliminary results, low eicosapentaenoic acid levels were linked with intracerebral hemorrhage during the COVID-19 pandemic. Low levels of eicosapentaenoic acid might be an endogenous surrogate marker for intracerebral hemorrhage during the COVID-19 pandemic.

Apostichopus japonicus and Parastichopus californicus are two of the most important and profitable com. sea cucumbers along the North Pacific coast. This study compared the body wall production rate (BWPR), proximate composition, amino acid, fatty acid, trace element and vitamin composition, and nonspecific immune enzyme activities of A. japonicus and P. californicus cultured in an artificial pond. The BWPR, crude fat and ash content in the body walls of A. japonicus and P. californicus showed remarkable differences (P < 0.05). For the 18 amino acids tested, differences in the contents of 15 were significant (P < 0.05) between the two species, except for threonine, methionine and histidine, and their first limiting amino acids were both methionine+cysteine. There were seven saturated and ten unsaturated fatty acids in their body walls, and except for 18:1 and 20:1, the content differences of the other 15 fatty acids were all significant (P < 0.05). Furthermore, between the two sea cucumbers, differences in the content of seven trace elements (Cu, Fe, Mn, Zn, Cr, Ni, Se) and six vitamins (B1, B3, B5, B9, C, E) were significant (P < 0.05). The activities of superoxide dismutase (SOD), catalase (CAT), acid phosphatase (ACP) and alk. phosphatase (AKP) also showed distinct differences (P < 0.05). There are greater differences in the biochem. compositions and contents between A. japonicus and P. californicus, each with its own unique quality advantages. In this work, a kinetics-integrated CFD model based on coupling computational fluid dynamics (CFD) and biokinetic was developed to guide the industrial scale-up for fermentation of docosahexaenoic acid (DHA). Two-substrate biokinetic equations for a 5 L bioreactor with oxygen and nitrogen source as limiting substrates were established, which were further coupled with CFD to simulate the biomass growth and lipid accumulation as well as flow field environment in the fermentation process. The coupled CFD-biokinetic model can accurately reflect the changes of each relevant variable in the 50 L bioreactor under different operating conditions. The optimal working conditions of a 35 m3 bioreactor were predicted by this model and have secured the one-step scale-up of DHA fermentation, resulting in a biomass, lipid concentration, and DHA content in lipids of 99.2 g/L, 55.7 g/L, and 52.5%, resp. This study thus provides an effective strategy for rapid industrial scale-up of DHA production

The peroxisomal very long chain fatty acid (VLCFA) transporter ABCD1 is central to fatty acid catabolism and lipid biosynthesis. Its dysfunction underlies toxic cytosolic accumulation of VLCFAs, progressive demyelination, and neurol. impairments including X-linked adrenoleukodystrophy (X-ALD). We present cryo-EM structures of ABCD1 in phospholipid nanodiscs in a nucleotide bound conformation open to the peroxisomal lumen and an inward facing conformation open to the cytosol at up to 3.5 ? resolution, revealing details of its transmembrane cavity and ATP dependent conformational spectrum. We identify features distinguishing ABCD1 from its closest homologs and show that CoA (CoA) esters of VLCFAs modulate ABCD1 activity in a species dependent manner. Our data suggest a transport mechanism where the CoA moieties of VLCFA-CoAs enter the hydrophilic transmembrane domain while the acyl chains extend out into the surrounding membrane bilayer. The structures help rationalize disease causing mutations and may aid ABCD1 targeted structure-based drug design.

It is essential to formulate artificial diets for Cyclina sinensis to satisfy the requirements of industrial aquaculture. We evaluated the effects of different dietary sources on the growth, survival, nutritional components, digestive performance and nonspecific immunity of C. sinensis. The diets of the three exptl. groups were as follows: natural bait (control group, CG), fermented Enteromorpha prolifea (EP) compound diet (Diet #1), and fermented Pyropia yezoensis (PY) compound diet (Diet #2). Feeding these diets did not affect the individuals final weight, specific growth rate and survival rate. The total amino acid and total monounsaturated fatty acid of individuals fed with Diet #1 were significantly higher than those provided with the other diets (p < 0.05). There were no significant differences in the activities of ¦Á-amylase, lipase, trypsin, total superoxide dismutase, lysozyme and acid phosphatase in the clam fed with different diets. In conclusion, the two fermented seaweed compound diets prepared herein can replace natural bait and meet the growth and developmental needs of C. sinensis. The fermented EP compound diet can improve the nutritional quality of the clam. This study lays a preliminary foundation for developing new artificial diets for clams and elucidates the further utilization of low-value seaweed resources.

Effects of vacuum packaging (VP) on quality changes of cooked and peeled harpiosquillid mantis shrimp (HMS) during storage at 4¡ãC of 15 days were investigated. VP inhibited bacterial growth as ascertained by the lower rate of increases in total viable and psychrotrophic bacteria counts as well as other pathogenic and spoilage bacteria counts. VP samples had lower total volatile base and trimethylamine contents as compared to the sample packed in air (AP). Clostridium perfringens was undetectable in both samples. During storage, VP samples possessed lower pH than AP counterpart. Exudate and TCA-soluble peptide (TSP) contents of sample increased during the storage, regardless of packaging atmospheres. Fatty acid (FA) content, especially PUFA and MUFA, was decreased after 9 and 15 days of storage for AP and VP samples, resp. The cooked and peeled HMS contained 32 volatile organic compounds, consisting of several aldehydes and alcs. Carnobacterium and Pseudomonas were mainly involved in spoilage in samples as confirmed by next generation sequencing.
