A practical and efficient potassium tert-butoxide (KOtBu)-facilitated amination of carboxylic acids with N,N-dimethylamine was described. In the presence of catalytic amount of KOtBu, a variety of aliphatic and aromatic carboxylic acids were transformed to N,N-dimethylamides using DMF as the dimethylamine reagent with the assistance of trimethylacetic anhydride. The applicability of this protocol was demonstrated by late-stage dimethylamidation of complex drug mols. A plausible reaction mechanism involving KOtBu-facilitated in-situ amine generation from formamide decomposition and anhydride-mediated condensation was proposed on the basis of mechanistic investigations.

Here, Authors report a photoinduced palladium-catalyzed hydrogen atom transfer triggered 1,2-difunctionalization of conjugated dienes. Without the employment of exogeneous photosensitizers and external oxidants, the cascade reaction realized the integration of remote functionalization of various C(sp3)-H bonds and selective difunctionalization of 1,3-dienes with 100% atom efficiency, allowing for the synthesis of structurally diverse amides with up to 90% yields. Given the prevalence of amides in pharmaceuticals and natural products, the current protocol has provided an efficient means to access highly functionalized amides from readily available carboxylic acid derivatives and 1,3-dienes.

A lead (Pb) exposure can alter the composition and metabolites of gut microbiota. However, few studies investigated this association in the children. A total of 551 children aged 3-7 years were recruited from Guiyu (the e-waste dismantling area) and Haojiang (the reference area). There were finally 70 subjects met the inclusive criteria. Blood and urinary Pb concentrations were detected by GFAAS and ICP-MS techniques. The microbiota and metabolites were measured in stool samples using 16 S rRNA MiSeq sequencing technol. and gas chromatog.-mass spectrometry (GC-MS), resp. Average Pb concentrations in the blood and urine of children were higher in Guiyu than in Haojiang. There were 58 kinds of differential genera and 19 types of discrepant metabolites between the two groups, and wide and significant correlations were found between them. Exposure to Pb caused the most significant differences in microbiota, metabolites, and phys. development parameters between the two groups in terms of microbiota, metabolites, and phys. development indicators. Sphingolipid metabolism and ion transport may also be altered by Pb exposure. Exposure to Pb is associated with significant alterations in the gut microbiota and metabolome in children. More research is needed to confirm the findings of this study.

In the present research, developed a new series of acetylphenol-based acyl thioureas I [R = t-Bu, 2-MeC6H4, 4-ClC6H4, etc.] that could potentially provide a new template for drug candidates to inhibit urease enzyme. Newly synthesized compounds I were evaluated for urease inhibitory strength using thiourea as a pos. control. In vitro inhibitory results revealed that all the tested compounds were significantly potent than the standard drug. The most active lead compound I [R =3-ClC6H4] competitively inhibited the enzyme and displayed an IC50 value of 0.054 ¡À 0.002¦ÌM, a ?413-fold strong inhibitory potential than thiourea (IC50 = 22.3 ¡À 0.031¦ÌM). Various insightful structure-activity relationships were developed showing the key structural requirements for potent inhibitory effects. Mol. docking anal. of compound I [R =3-ClC6H4] inside the active pocket of urease suggested several important interactions with amino acid residues such as ILE411, MET637, ARG439, GLN635, ALA636 and ALA440. Finally, pharmacokinetic properties suggested that the tested derivatives were safe to develop as low-mol.-weight drugs to treat ureolytic bacterial infections.

A practical route for the synthesis of valuable 3-aryl anthranils from readily available anthranils and simple arenes by using the classical electrophilic aromatic substitution (EAS) strategy was reported. This transformation goes through an electrophilic substitution and rearomatisation sequence by employing Tf2O as an effective activator. A wide range of arenes were compatible in this transformation, delivering various structurally diversified 3-aryl anthranils in good yields and high regioselectivity. In addition, a variety of readily available feedstocks such as olefins, alkenyl triflates, silyl enolethers, carbonyl compounds, thiophenols and thiols could also participate in the reaction to achieve the C3 alkenylation, alkylation and thioetherification of anthranils. Of note, the synthesized 3-aryl anthranils proved to be a highly robust platform to access a series of biol. active compounds, drug derivatives and organic optoelectronic materials.

The increments in conversion unattainable under external heating condition are realized and the extension to the mild oxidation of thioethers and olefins into sulfoxides and epoxides with acceptable catalytic recyclability and structural stability are demonstrated. All results from the detailed characterizations figure out the applicability of the NIR photothermal conversion to the catalytic reactions with similar catalytic centers.
