In this study, panaxadiol (PD) derivs, I [R1 = 2-Me, 3-O2N, 3,4,5-trimethoxy, etc.] and II [R2 = H, AcO] were designed and synthesized by the reaction of thiadiazoles with PD. The identity of all final products I and II was elucidated by 1H, 13C NMR, HR-MS. The cytotoxicity activities of the derivatives I and II were evaluated against four cancer cell lines using the MTT assay which revealed they indicated excellent anticancer activity. Among these compounds, II [R2 = H] had the most significant cytotoxicity and distinctly inhibited the growth of a variety of tumor cells. Further studies showed that compound II [R2 = H] could decrease the expression levels of CDKs protein family and Cyclin D1 in A549, suggesting that compound II [R2 = H] could block the cell cycle. To prove the above conclusions, the binding sites of the two proteins were simulated by mol. docking method. The results demonstrated that the docking scores of II [R2 = H] and the two cyclins are higher than that of PD. Based on the complex of the target protein and ligand, it was speculated that the enhanced antiproliferative activity may be related to the increased hydrogen bonding interactions. Therefore, these data provide a reference that compound II [R2 = H] could be a promising lead drug for cancer treatment.

This method afforded aromatic carbonyl compounds under TBHP via selective oxidative cleavage of the C-C bond in ¦Á,¦Â-epoxy ketones. Aromatic acid came from the aroyl section, and aromatic aldehyde came from the other aromatic group. TBHP acted as a free radical initiator and oxidant. The reaction within the solvent went through a ring-opening addition, cleavage of the C-C bond in the ethylene oxide section, and oxidation, affording the target compounds in moderate to good yields. The HPLC yield of aromatic aldehyde was up to 91%. The HPLC yield of aromatic acid was up to 99%. The reaction under solvent-free conditions gave two kinds of aromatic acids coming from different moieties of ¦Á,¦Â-epoxy ketone via the further oxidation of aromatic aldehyde. The substituent effect was discussed, and the reaction mechanism was proposed. This method allowed the reaction to occur in a simple system metal-free.

The obtained catalysts were utilized for the selective oxidation of alcs. to high value-added esters and carboxylic acids with air as the oxygen source and displayed wide applicability for the oxidation of both aromatic and aliphatic alcs. Based on the controlled experiments, the protective effect of N-doped carbon structure and the synergistic effect between Co core and Co3O4 species guaranteed the high reaction conversion and selectivity. Benefitting from heterogeneity and magnetism of the catalyst, it can be easily recycled and reused for long-term stability. Reasonable mechanisms of selective oxidation reaction were proposed through EPR anal. and controlled experiments The present work provides a facile strategy for potential large-scale preparation of heterogeneous catalyst for sustainable and green catalytic transformations.

A library of twenty-four 2′-aryl-2,5′-bibenzoxazoles I [R1 = H, Me, OH; R2 = H, Me; R3 = H, Me, F, Cl, Br; R4 = H, Me; R5 = H, Cl] had been synthesized from 2-arylbenzoxazole-5-carboxylic acids which in turn were obtained by hydrolysis of Me 2-arylbenzoxazole-5-carboxylates in excellent yields. These carboxylates were prepared form Me 3-amino-4-hydroxybenzoate and readily available various benzoic acids. The bibenzoxazoles were characterized using NMR, FT-IR and HRMS techniques and the structure of compound I [R1 = OH; R2 = R3 = R4 = R5 = H] was established by X-ray crystallog. All the synthesized compounds were screened for cytotoxicity (in vitro) against cervical cancer (HeLa) and hepato-carcinoma (HepG2) cell lines and non-cancerous (HEK293) cell line. Compounds, I [R1 = H, Me, OH; R2 = H, Me; R3 = H, F, Br; R4 = R5 = H] showed good activity against HeLa cancer lines having the IC50 values 4.787, 9.611 and 10.68¦ÌM, resp.

An innovative 1,5-HAT cascade strategy was advanced for the nickel-catalyzed distal arylation via cross-electrophile coupling. Through specific migration, the remote C(sp3)-H bond was regioselectively activated, and Ar-I as the available electrophile is used for the construction of the C(sp3)-C(sp2) bond. This method also has broad applicability for benzylic and aliphatic N-fluorocarboxamides with yields up to 80%. Furthermore, a series of control experiments demonstrated that this reaction was probably initiated by a radical process.
