Pathogenic bacteria cause disease outbreaks and threaten human health, prompting the research on advanced detection assays. Herein, we developed a selective mol. imprinted aptasensor for sensitive and prompt quantitation of Staphylococcus aureus (S. aureus) bacteria. The aptasensor was constructed by immobilization of aptamer on gold nanoparticles modified magnetic nanoparticles (apt-AuNPs@ Fe3O4). A functional monomer (o-phenylenediamine, o-phen) was electro-polymerized on the surface of the as-synthesized nanocomposite in the presence of a template (S. aureus). After removing S. aureus, the formed imprinted sites were available to extract pathogenic bacteria from complicated matrixes. The surface morphol. of the as-fabricated nanocomposites was characterized using different spectroscopic and electrochem. methods. Moreover, we thoroughly evaluated factors affecting the synthesis and determination procedures. The mol. imprinted aptasensor exhibited a wide linear range of 101-107 CFU mL-1 with a Limit of Detection, LOD (signal to noise = 3) of 1 CFU mL-1. The aptasensor detected S. aureus in milk, conduit water, and apple juice samples with good recoveries % and satisfactory relative standard deviations (RSDs %) values.

In this study, the techno-functional and phys. attributes of various com. available pulse protein isolates were compared with those of soy protein isolate to evaluate their potential application as replacer for soy proteins in liquid and solid food formulations such as meat and dairy analogs. The water holding capacity, oil holding capacity, gelation properties, emulsifying properties, and color of faba bean (FPI), pea (PPI), lentil (LPI), and soy (SPI) protein isolates were therefore measured. SPI had a significantly higher water holding capacity (7.6 g/g) than the pulse protein isolates (2.2-5.1 g/g), but similar oil holding capacities were identified for SPI, FPI, and LPI. Especially LPI was more effective in droplet size reduction already at low concentrations (0.1-0.2%) compared to all other tested proteins, whereas FPI showed similar droplet size reduction efficiencies than SPI at higher concentrations (2-5%). Nevertheless, all of the plant proteins were capable of forming relatively small oil droplets (D32 = 1-3¦Ìm) at a protein-to-oil ratio of 1:10. Finally, FPI exhibited similar min. gelling concentrations to SPI, and especially LPI had a low coloring potential.

Sorghum (Sorghum bicolor L. (Moench)) is the world’s fifth economically most important cereal and is a staple particularly in the semi-arid tropics of Africa and Asia. Genetic gains in this crop can benefit from wild relatives such as Sorghum halepense. Genome sequences including those from this wild species can boost the study of genome-wide and intraspecific variation for dissecting the genetic basis and improving important traits in sorghum. The whole-genome resequencing carried out in this work on a panel of 172 populations of S. bicolor and S. bicolor x S. halepense (SbxSh) advanced lines generated a total of 567,046,841 SNPs, 91,825,474 indels, 1,532,171 SVs, and 4,973,961 CNVs. Clearly, SbxSh accumulated more variants and mutations with powerful effects on genetic differentiation. A total of 5,548 genes private to SbxSh mapped to biol. process GO enrichment terms; 34 of these genes mapped to root system development (GO: 0022622). Two of the root specific genes i.e., ROOT PRIMORDIUM DEFECTIVE 1 (RPD1; GeneID: 8054879) and RETARDED ROOT GROWTH (RRG, GeneID: 8072111), were found to exert direct effect on root growth and development. This is the first report on whole-genome resequencing of a sorghum panel that includes S. halepense genome. Mining the private variants and genes of this wild species can provide insights capable of boosting sorghum genetic improvement, particularly the perenniality trait that is compliant with agroecol. practices, sustainable agriculture, and climate change resilience.

Bradyrhizobium japonicum strain SEMIA 5079 (= CPAC 15) is a nitrogen-fixing symbiont of soybean broadly used in com. inoculants in Brazil. Its genome has about 50% of hypothetical (HP) protein-coding genes, many in the symbiosis island, raising questions about their putative role on the biol. nitrogen fixation (BNF) process. This study aimed to infer functional roles to 15 HP genes localized in the symbiosis island of SEMIA 5079, and to analyze their expression in the presence of a nod-gene inducer. A workflow of bioinformatics tools/databases was established and allowed the functional annotation of the HP genes. Most were enzymes, including transferases in the biosynthetic pathways of cobalamin, amino acids and secondary metabolites that may help in saprophytic ability and stress tolerance, and hydrolases, that may be important for competitiveness, plant infection, and stress tolerance. Putative roles for other enzymes and transporters identified are discussed. Some HP proteins were specific to the genus Bradyrhizobium, others to specific host legumes, and the anal. of orthologues helped to predict roles in BNF. All 15 HP genes were induced by genistein and high induction was confirmed in five of them, suggesting major roles in the BNF process.

Both hyperthyroidism and elevated plasma branched chain amino acids (BCAA) are associated with insulin resistance. BCAA utilization and clearance relative to thyroid status changes remains unclear. We investigate amino acids changes, specifically BCAA, during the transition from hyperthyroidism to euthyroidism, and the impact of active brown adipose tissue (BAT) on the metabolic effects of BCAA. Newly diagnosed Graves disease participants were recruited. Hyperthyroidism was treated via a titration dosing regimen of thionamide anti-thyroid drug to establish euthyroidism over 12-24 wk. All underwent energy expenditure (EE) measurement within a chamber calorimeter, 18F-fluorodeoxyglucose (18F-FDG) positron-emission tomog./magnetic resonance (PET/MR) imaging and plasma amino acids measurement during hyperthyroidism and euthyroidism. PET BAT maximum standardized uptake value (SUVmax), SUVmean and MR supraclavicular fat fraction (FF) quantified BAT activity. Twenty-two patients completed the study. Plasma BCAA level was significantly reduced in BAT-pos. but not in BAT-neg. patients during the transition from hyperthyroidism to euthyroidism. Plasma valine but not leucine and isoleucine correlated pos. with insulin and HOMA-IR in hyperthyroidism. Plasma valine, leucine and isoleucine correlated with insulin and HOMA-IR in euthyroidism. Plasma valine correlated with insulin and HOMA-IR in BAT-neg. but not in BAT-pos. participants in both hyperthyroid and euthyroid state. However, the change (i.e. decrease) in plasma valine concentration from hyperthyroid to euthyroid state was affected by BAT-status. BAT utilizes and promotes BCAA plasma clearance from hyperthyroid to euthyroid state. Active BAT can potentially reduce circulating BCAA and may help to ameliorate insulin resistance and improve metabolic health. Leftover dough is a starch-rich food processing waste of Chinese steamed bread. Leftover dough hydrolyzates enriched with glucose and amino acids were used to cultivate the marine microalga Isochrysis galbana to produce docosahexaenoic acid (DHA) under CO2 enrichment. Isochrysis galbana could use mixed carbon sources (CO2, glucose, and amino acids) synchronously to grow and accumulate DHA. Cell growth, the uptake of glucose and amino acids, and DHA production were significantly affected by CO2 enrichment. The maximum biomass concentration of 3.85 g L-1 was achieved with 3% CO2. And the maximum DHA yield was 65.5 mg L-1 d-1. To enhance DHA production, a two-stage cultivation strategy was successfully developed by this work. The maximum DHA yield of the two-stage culture was elevated by 2.3-fold. It is feasible to produce DHA by Isochrysis galbana using leftover dough under CO2 enrichment.

Resource utilization of chrome shavings (CS) has attracted a lot of attention from scientists and technologists in leather industry. Especially, the collagen hydrolyzates extracted from CS are expected to find potential application values in agricultural field. However, there is no biotoxicity anal. of collagen hydrolyzates from CS. Herein, the collagen hydrolyzates with different mol. weights were produced from CS by three hydrolysis dechroming methods including alk. hydrolysis, enzymic hydrolysis and alk.-enzymic synergistic hydrolysis, and the optimal hydrolysis process of CS was designed and conducted. To evaluate their toxicity, the three collagen hydrolyzates were formulated into a nutrient solution for zebrafish development. The obtained results indicated that the hydrolyzates with low concentrations (less than 0.6 mg/mL) were safe and could promote the development for zebrafish embryos. Furthermore, the three collagen hydrolyzates were utilized as organic nitrogen sources and formulated into amino acid water-soluble fertilizers (AAWSF) including alk. type fertilizer (OH), enzymic type fertilizer (M) and alk.-enzymic type fertilizer (OH-M) for the early soilless seeding cultivation of wheat, soybean and rapeseed. It is worth mentioning that the chromium contents in the prepared AAWSF were less than 10 mg/kg, which is far less than the limit value in the standard (China, 50 mg/kg). The growth and development of seedlings (germination rate, plant height, fresh weight of leaves, soluble sugar content and chlorophyll content) were investigated. The corresponding results showed that the growth of seedlings watered with AAWSF was better compared with the other treatments, and the OH-M fertilizer had the best promoting effect on the seedlings growth and development, followed by the M and OH fertilizers. The safe toxicity assessment of the collagen hydrolyzates will expand their application scope, and the use of collagen hydrolyzates extracted from CS for seedlings growth also provides an effective and reasonable way to deal with the chromium-containing leather solid waste, which is an effective way to realize its resource utilization.

Musculoskeletal pain conditions (MPs) are a widespread public problem that can affect 13.5% to 47% of the total population. Dietary changes can have strong effects on persons health; for instance, Sulfur amino acids (SAAs) can act as a precursor of neurotransmitters, antioxidative metabolic intermediates, such as glutathione, impact inflammation, and play a role in severity and frequency of MPs. We evaluated the relationship between dietary SAAs intake with severity and frequency of pain in patients with MPs. This cross-sectional study consisted of 175 men and woman. Anthropometric measurements and pain assessments were conducted via questionnaires. Dietary data were collected using 7 days 24-h recall. ANOVA and Spearman correlation coefficients were used to examine the relationship and correlation, resp., between exposure and outcome variables. There was a significant correlation between age, weight, waist circumference (WC), waist circumference to height (WHtR), body mass index (BMI), and severity and frequency of MPs among women. There was a correlation between age and severity of pain in men. The present study highlights a pos. association between the dietary SAAs and severity of pain, even after adjusting for confounding variables.

Abstract: Background: Colistin is a polymyxin antibiotic which has been used for treatment of Gram-neg. infections, but it was withdrawn due to its nephrotoxicity. However, colistin has gained its popularity in recent years due to the reemergence of multidrug resistant Gram-neg. infections and drug-induced toxicity is considered as the main obstacle for using this valuable antibiotic. Results: In total, 30 articles, including 29 animal studies and one clin. trial were included in this study. These compounds, including aged black garlic extract, albumin fragments, alpha lipoic acid, astaxanthin, baicalein, chrysin, cilastatin, colchicine, curcumin, cytochrome c, dexmedetomidine, gelofusine, grape seed proanthocyanidin extract, hesperidin, luteolin, lycopene, melatonin, methionine, N-acetylcysteine, silymarin, taurine, vitamin C, and vitamin E exhibited beneficial effects in most of the published works. Conclusions: In this review, the authors have attempted to review the available literature on the use of several compounds for prevention or attenuation of colistin-induced nephrotoxicity. Most of the studied compounds were potent antioxidants, and it seems that using antioxidants concomitantly can have a protective effect during the colistin exposure.

Toxoplasma gondii commonly infects humans and while most infections are controlled by the immune response, currently approved drugs are not capable of clearing chronic infection in humans. Hence, approx. one third of the world¡äs human population is at risk of reactivation, potentially leading to severe sequelae. To identify new candidates for treating chronic infection, we investigated a series of compounds derived from diversity-oriented synthesis. Bicyclic azetidines are potent low nanomolar inhibitors of phenylalanine tRNA synthetase (PheRS) in T. gondii, with excellent selectivity. Biochem. and genetic studies validate PheRS as the primary target of bicyclic azetidines in T. gondii, providing a structural basis for rational design of improved analogs. Favorable pharmacokinetic properties of a lead compound provide excellent protection from acute infection and partial protection from chronic infection in an immunocompromised mouse model of toxoplasmosis. Collectively, PheRS inhibitors of the bicyclic azetidine series offer promise for treatment of chronic toxoplasmosis.
