Steroidal thiazoles I (R = NH2, NHMe, NHPh, NMe2, Ph, Me, CH2CO2Me) have been synthesized. The starting bromo ketone II (R1 = Br) was prepared by bromination of pregnen-20-ones II (R1 = H) with copper (II) bromide, and was used for synthesis of the thiazole derivatives employing the Hantzsch reaction. Preliminary biol. evaluation of thiazoles I and III against the P388 lymphocytic leukemia cell showed growth inhibition values of ED50 2.9 and 7 ¦Ìg/mL for thiazoles III and I (R = NH2), resp. Other I were less active.

In presence of some mol. moieties, ¦¤fGcorr¦È may increase or decrease the value of ¦¤fGelem¦È, depending on the intermol. interactions. The exptl. root-mean-square error (RMSE) of the novel correlation (22.7 kJ mol-1) is quite good. For some energetic compounds, where the computed values of two complex models of the quant. structure-property relationship (QSPR) theory were available, the exptl. RMSE developed by the new method is smaller than the values obtained by QSPR method.

The selective CuII-chelator, triethylenetetramine (TETA), is undergoing clin. trials for the treatment of heart failure in patients with diabetes. Recently, the authors showed that 2 acetylated metabolites, N1-acetyltriethylenetetramine (MAT) and N1,N10-diacetyltriethylenetetramine (DAT), are formed in humans following oral TETA administration. Thus, it became necessary to determine whether the N-acetyltransferase (NAT) 2 phenotype has any effects on the pharmacol. properties and safety profile of TETA. Twelve fast and 12 slow NAT2-phenotype healthy participants were recruited. After oral drug administration, the authors collected plasma and urine samples, measured plasma concentrations of TETA and its 2 metabolites along with concomitant urinary copper concentrations, and performed safety tests. They present, for the first time, the complete 24-h pharmacokinetic profiles of TETA, MAT, and DAT in humans. There was no evidence for clear-cut differences in pharmacokinetic profiles between fast and slow acetylators. Pharmacodynamic anal. showed no significant differences in cupruresis between the 2 NAT2 phenotypes. Safety results were consistent with TETA being well tolerated, and no significant differences in safety profiles were observed between the 2 phenotypes. Based on these data, NAT2 phenotype does not affect TETA’s pharmacokinetic, pharmacodynamic, or safety profiles. TETA may be acetylated via an alternative mechanism, such as that catalyzed by spermidine/spermine N1-acetyltransferase.

PM2.5 is a widely regarded major air pollutant due to its adverse health effect and intimate relationship with the climate system. This work characterized the chem. components (e.g., organic and elemental C [OC, EC], water soluble ionic species [WSIS], polycyclic aromatic hydrocarbons [PAH]) in PM2.5 collected over Doi-Inthanon in Chiang-Mai, the highest mountain in Thailand. All samples (n = 50) were collected by MiniVolTM portable air samplers Mar. 2017-Mar. 2018. The average PM2.5 concentration was 100 ¡À 48.6¦Ìg/m3. The OC:EC ratio was 6.8 ¡À 3.0; the decreasing order of WSIS concentrations was: SO42- > Na+ > Ca2+ > NH4+ > NO3 > K+ > Cl > NO2 > Mg2+ > F. Total concentrations of nineteen PAH were defined as the sum of Ace, Fl, Phe, Ant, Fluo, Pyr, B[a]A, Chry, B[b]F, B[k]F, B[a]F, B[e]P, B[a]P, Per, Ind, B[g,h,i]P, D[a,h]A, Cor, and D[a,e]P. The total PAH concentration 2.361 ¡À 2.154¦Ìg/m3. Principal component anal. (PCA) highlighted the importance of vehicle exhaust, biomass burning, diesel emissions, sea-salt aerosols, and fertilizer volatilization as the five dominant potential sources, accounting for 51.6, 16.2, 10.6, 5.20, and 3.70% of total variance, resp. The remaining 12.7% variance is associated with unidentified local and regional sources, e.g., incinerators, joss paper/incense burning, domestic cooking. PCA source estimation results underlined the importance of vehicle exhaust as the major PM2.5 contributor in ambient air over Don-Inthanon, Chiang-Mai Province. It is crucial to emphasize the effect of agricultural waste burning, fossil fuel combustion, coal combustion, and forest fires on OC, EC, and WSIS concentration variations was not negligible.
