By using cheap, easy-to-handle, and low-toxic 4-cyanopyridine as the cyanating source, cyanation of various aromatic amides afforded an assortment of aryl nitriles including bioactive drugs and organic luminescent mols. in good yields. The reaction exhibited wide substrate scope, good functional group tolerance, and unique selectivity that were complementary to traditional methods. Moreover, two key nickel complexes formed by oxidative addition to each substrate are obtained and determined by X-ray crystallog., which gave strong support for the mechanism elucidations.

A series of novel quinazolinone hydrazide derivatives were designed and synthesized as EGFR inhibitors. The results indicated that most of the aimed compounds had potential anti-tumor cell proliferation and EGFR inhibitory activities. In the comprehensive anal. of all the tested compounds, the target compound 9c showed the best anti-tumor cell proliferation activity, (IC50=1.31 ¦ÌM for MCF-7, IC50=1.89 ¦ÌM for HepG2, IC50=2.10 ¦ÌM for SGC), and IC50=0.59 ¦ÌM for the EGFR inhibitory activity. Docking results showed that compound 9c could ideally insert the active site and interact with the critical amino acid residues (Val702, Lys721, Met769, Asp831) in the active site.

In the presence of catalytic amount of KOtBu, a variety of aliphatic and aromatic carboxylic acids were transformed to N,N-dimethylamides using DMF as the dimethylamine reagent with the assistance of trimethylacetic anhydride. The applicability of this protocol was demonstrated by late-stage dimethylamidation of complex drug mols. A plausible reaction mechanism involving KOtBu-facilitated in-situ amine generation from formamide decomposition and anhydride-mediated condensation was proposed on the basis of mechanistic investigations.

Fish is favored by consumers, while amino acids and fatty acids are the main nutrients of muscle. At present, it has been found that the gut microbial community may be involved in the regulation of host material anabolism. Juvenile and adult bighead carp (A. nobilis) from Chagan lake and Xinlicheng reservoir were selected, and divided into four groups to compare the differences of gut microbial communities, free amino acid and fatty acids in muscle. The results showed that fish in different lakes or ages contained specific microbiota, the gut microbial structure was similar, but the microbial content was significantly different. Gut microbial abundance of juvenile fish in Chagan lake was significantly higher than that of other groups. Phylum level anal. Proteobacteria was the dominant gut bacteria of fish in both adult and juvenile fish from two sep. lakes. Actinobacteria was another dominant bacterial phylum in juvenile fish in both lakes. Contents of free amino acids and fatty acids in muscle were detected, and the relationships between them and gut microbial communities were analyzed. Bighead carp grew from juvenile to adult, Actinobacteria abundance decreased (P < 0.05) and Proteobacteria increased (P < 0.05). Proteobacteria was pos. correlated with the contents of Thr, Lys, Pro, Asp, Gly and Glu, Actinobacteria was neg. correlated with Met and His. Meanwhile, EPA and DHA were pos. correlated with Proteobacteria, EPA and DHA were not significantly associated with Actinobacteria. It was speculated that the contents of free amino acids and fatty acids in muscle might be affected by the difference of gut microbiota, thus affecting the taste and nutritional quality.

Here, Authors report a photoinduced palladium-catalyzed hydrogen atom transfer triggered 1,2-difunctionalization of conjugated dienes. Without the employment of exogeneous photosensitizers and external oxidants, the cascade reaction realized the integration of remote functionalization of various C(sp3)-H bonds and selective difunctionalization of 1,3-dienes with 100% atom efficiency, allowing for the synthesis of structurally diverse amides with up to 90% yields. Given the prevalence of amides in pharmaceuticals and natural products, the current protocol has provided an efficient means to access highly functionalized amides from readily available carboxylic acid derivatives and 1,3-dienes.
