4-Hydroxyphenylpyruvate dioxygenase (EC 1.13.11.27, HPPD) is an important target for new bleaching herbicides discovery. To search for novel HPPD inhibitors, a series of 2-(aryloxyacetyl)cyclohexane-1,3-diones were rationally designed and synthesized by an efficient one-pot procedure using of N,N’-Carbonyldiimidazole (CDI), triethylamine, and acetone cyanohydrin in CH2Cl2. A total of 58 triketone compounds were prepared in good to excellent yields. Some of the synthesized compounds displayed potent Arabidopsis thaliana HPPD (AtHPPD) inhibitory activity. 2-(2-((1-Bromonaphthalen-2-yl)oxy)acetyl)-3-hydroxycyclohex-2-en-1-one (I) displayed high, broad-spectrum and post-emergent herbicidal activity at the rate of 37.5-150 g ai/ha, nearly as potent as mesotrione against some weeds. Furthermore, I showed good crop safety against maize and canola at the rate of 150 g ai/ha, indicating that I might have the potential as herbicide for the weed control in maize and canola fields. I is the first HPPD inhibitor showing good crop safety towards canola.

The method provides a novel approach to access valuable carbonyls from benzylic methylenes and benzylic alcs. in moderate to excellent yields (up to 93 %).

The COVID-19 pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continually poses serious threats to global public health. The main protease (Mpro) of SARS-CoV-2 plays a central role in viral replication. We designed and synthesized 32 new bicycloproline-containing Mpro inhibitors derived from either boceprevir or telaprevir, both of which are approved antivirals. All compounds inhibited SARS-CoV-2 Mpro activity in vitro, with 50% inhibitory concentration values ranging from 7.6 to 748.5 nM. The cocrystal structure of Mpro in complex with MI-23, one of the most potent compounds, revealed its interaction mode. Two compounds (MI-09 and MI-30) showed excellent antiviral activity in cell-based assays. In a transgenic mouse model of SARS-CoV-2 infection, oral or i.p. treatment with MI-09 or MI-30 significantly reduced lung viral loads and lung lesions. Both also displayed good pharmacokinetic properties and safety in rats.

In order to find new herbicidal leading compounds, a series of N-(4-chloro-2-fluoro-5-substituted phenyl)isoindole-1,3-dione derivatives were designed and synthesized via the strategy of linking active sub-structures which are chlorophthalim and phenoxy carboxylic ester. The target compounds were synthesized from starting material 2-chloro-4-fluoro-5-nitrophenol in four steps, and their structures were confirmed by 1H NMR, 13C NMR and HRMS. The preliminary bioassay results indicated that most compounds exhibited excellent herbicidal activities at 1,500 g/ha in both pre- and post-emergence treatments against Abutilon theophrasti and Amaranthus retroflexus. On the basis of advanced screening tests, compound 6d exhibited 100% inhibitory effect against Abutilon theophrasti and Amaranthus retroflexus at 750 g/ha, compounds 7a and 7i were much more effective against Abutilon theophrasti than com. acifluorfen with 80%-86% inhibitory activity at 22.5 g/ha.
