Fish is favored by consumers, while amino acids and fatty acids are the main nutrients of muscle. At present, it has been found that the gut microbial community may be involved in the regulation of host material anabolism. Juvenile and adult bighead carp (A. nobilis) from Chagan lake and Xinlicheng reservoir were selected, and divided into four groups to compare the differences of gut microbial communities, free amino acid and fatty acids in muscle. The results showed that fish in different lakes or ages contained specific microbiota, the gut microbial structure was similar, but the microbial content was significantly different. Gut microbial abundance of juvenile fish in Chagan lake was significantly higher than that of other groups. Phylum level anal. Proteobacteria was the dominant gut bacteria of fish in both adult and juvenile fish from two sep. lakes. Actinobacteria was another dominant bacterial phylum in juvenile fish in both lakes. Contents of free amino acids and fatty acids in muscle were detected, and the relationships between them and gut microbial communities were analyzed. Bighead carp grew from juvenile to adult, Actinobacteria abundance decreased (P < 0.05) and Proteobacteria increased (P < 0.05). Proteobacteria was pos. correlated with the contents of Thr, Lys, Pro, Asp, Gly and Glu, Actinobacteria was neg. correlated with Met and His. Meanwhile, EPA and DHA were pos. correlated with Proteobacteria, EPA and DHA were not significantly associated with Actinobacteria. It was speculated that the contents of free amino acids and fatty acids in muscle might be affected by the difference of gut microbiota, thus affecting the taste and nutritional quality.

Allatostatins (ASTs), a family of insect neuropeptide, can inhibit juvenile hormone (JH) biosynthesis by the corpora allata (CA) in Diploptera punctata, and therefore be regarded as potential leads for the discovery of new insect growth regulators (IGRs). But several shortcomings, such as their sensitivity to peptidases and high cost, impeded their practical application in pest management. In order to discover new IGRs, one AST analog B1 (I) possessing non-peptide group was discovered with high ability to inhibit JH biosynthesis in vitro (IC50: 0.09 ¦Ìmol/L) in our previous studies. In the present work, two series of I analogs with different substituents on the N-terminus region were designed and synthesized. The result suggested that benzene showed better activity than other heterocycles, and the para-substitution on the benzene was beneficial for activity. Moreover, analogs with logP value over 2.0 exhibited good activity, which indicated the hydrophobicity is important to the bioactivity. Three dimension quant. structure-activity relationship (3D-QSAR) studies were performed to highlight the structural requirements of AST analogs, which demonstrated introduction of bulkier substituents on the N-terminus would increase the activity. Analog (II) (IC50: 0.08 ¦Ìmol/L) exhibited similar inhibitory activity to the lead B1, but its synthetic route was simpler than B1. Therefore, II could be used as a new lead compound for the discovery ecofriendly IGRs.
