A short and efficient sequence for the synthesis of a series of 4-(2-amino-5-thiazolyl)-pyrimidine-2-amines I [R1 = Ph, 4-MeOC6H4; R2 = MeNH, Me2N, H2N; Ph, 3-MeOC6H4, 3-(MeCO)C6H4] was developed. 1-Phenyl-2-(4-pyrimidinyl)ethanones, obtained via Weinreb’s methodol., were used in a Hantzsch thiazole cyclization reaction, followed by introduction of the aniline moieties via nucleophilic substitution.
