A new one-pot synthesis of 3-(1,2,3,4-tetrahydroisoquinolin-1-yl)-isoquinolin-1(2 H)-ones I (R1 = H, Cl, Br; R2 = TsCH2, C6H5CH2, CH2CO2OMe; R3 = Ph, 4-MeC6H4, etc.) by DEAD-promoted oxidative Ugi-Wittig reaction was developed. The sequential reactions of (2-carboxybenzyl)triphenylphosphonium salts II?Br– (R1 = H, Cl, Br), isocyanides R2NC, and N-aryl-1,2,3,4-tetrahydroisoquinolines III produced 3-(1,2,3,4-tetrahydroisoquinolin-1-yl)-isoquinolin-1(2 H)-ones I in moderate to good overall yields in the presence of DEAD and Et3N.
