A number of mevalonic acid analogs and their N,N’-dibenzylethylenediamine (I) salts were tested for their effect on the incorporation of mevalonate-2-14C into cholesterol by rat liver homogenates. In contrast to the mevalonic acid analogs, their I salts exhibited an inhibitory property which was found to reside in I itself. The site of action of I is at the level of the conversion of 7-dehydrocholesterol to cholesterol. I was moderately effective in lowering serum sterols in hypercholesterolemic rats. Mol. modifications of I have resulted in compounds of increased potency, showing activity in vitro at a concentration of 10-8M.
