The effects of polyamines on the melting of poly(riboadenylic acid) (I) and poly(ribouridylic acid) (II) were studied, where melting is defined as a transition in solution from the double (or triple) helical conformation into random coils. The absorbance was measured at 259 m¦Ì for several mixtures at 18¡ã in several concentrations of NaCl and Na citrate, NaCl, glycylglycine, and ethylenediamine solutions Absorbance observations indicated that a 1:2 complex of I:II was present in 0.1M NaCl + 0.01M glycylglycine buffer. The melting of the polymer mixtures was followed as dependent on the Na+ concentration in the solvent. Also, the melting was followed at 259 m¦Ì and 280 m¦Ì for the 1:1 mixture in 0.01M NaCl + 0.001 M Na citrate buffer. A mechanism is suggested for the phenomenon observed The effects of ethylenediamine dihydrochloride, 1,3-diaminopropane-2HCl, 1,4-diaminobutane-2HCl, 1,6-diaminohexane-2HCl, 1,8-diaminooctane-2HCl, and spermine-4HCl on the melting process were studied. Actinomycin D has no effect.
